چکیده

Background: Overweight and obesity represents a complex interaction of genetics, diet, metabolism, and physical activity. Abnormal or excessive body fat accumulation increases the risk of pathologies, such as heart disease, diabetes, and high blood pressure. Methods and results: In a previous paper, we investigated the influence of overweight individuals’ sera on in vitro biology of mesenchymal stromal cells, which contain a subpopulation of multipotent stem cells. This report extended the study by evaluating effects of sera collected from weight loss patients on stromal cultures. Results showed that an increase in fat mass induces physiological changes that negatively impact on stromal stem cell adipogenesis and osteogenesis with a bias toward adipocyte differentiation. This phenomenon occurred with sera from overweight patients and persisted also with sera of individuals that underwent loss of weight. Significant changes in obesity related cytokines were detected in sera of overweight and weight loss compared to healthy weight individuals. These changes may explain some of the biological effects detected on stromal stem cell functions. Effect of Lithium Chloride on Proliferation and Bone Differentiation of Rat Marrow-Derived Mesenchymal Stem Cells in Culture

Objective(s) It is believed that the mesenchymal stem cell (MSC) differentiation and proliferation are the results of activation of wnt signaling pathway. On the other hand, lithium chloride is reported to be able to activate this pathway. The objective of this study was to investigate the effect of lithium on in vitro proliferation and bone differentiation of marrow-derived MSC. Materials and ...

Ex vivo Expansion and Differentiation of Mesenchymal Stem Cells from Goat Bone Marrow

Objective(s) Mesenchymal stem cells (MSCs) from large animals as goat which is genetically more closely related to human have rarely been gained attentions. The present study tried to isolate and characterize MSCs from goat bone marrow. Materials and Methods Fibroblastic cells appeared in goat marrow cell culture were expanded through several subcultures. Passaged-3 cells were then different...

S-nitrosylation and MSC-mediated body composition

Mesenchymal stem cells (MSCs) are nonhematopoietic stromal cells that can differentiate into multiple mesodermal lineages including adipocytes, osteoblasts, chondrocytes and myocytes. Adult stem cell differentiation is controlled by activation of lineagespecific transcription factors, including peroxisome proliferator-activated receptor γ (PPARγ) and Runx2, two key transcription factors that go...

Multiple Myeloma Bone Marrow Mesenchymal Stromal Cells Inhibit CD8+ T Cell Function in a Process that May Implicate Fibroblast Activation Protein α

Background: Multiple myeloma (MM) is a malignant plasma cell proliferative disorder with limited immunotherapy treatment because of T cell dysfunction. Objective: To investigate the immunomodulatory function of bone marrow mesenchymal stromal cells (MM-BMSCs) on CD8+ T cells. Methods: Proliferation and cytotoxicity were detected by c...

Cyclic AMP signaling in bone marrow stromal cells has reciprocal effects on the ability of mesenchymal stem cells to differentiate into mature osteoblasts versus mature adipocytes

Stimulatory G protein-mediated cAMP signaling is intimately involved in skeletal homeostasis. However, limited information is available on the role of the cAMP signaling in regulating the differentiation of mesenchymal stem cells into mature osteoblasts and adipocytes. To investigate this, we treated primary mouse bone marrow stromal cells (BMSCs) with forskolin to stimulate cAMP signaling and ...
